For Automated Extraction of Genomic DNA from Cultured Cells and Amniotic Fluids

MagCore® Cultured Cells DNA Kit can be used to extract DNA from up to 5 x 106 cells via MagCore® automated extraction instruments. Different protocols are optimized for samples of cell suspensions, cells grown in a monolayer, and amniotic fluid from trypsinization to DNA elution. Our programs and protocols ensure maximal efficacy and efficiency in performance for the specific type of sample used.

    Table 1. Consistency in yield and reproducibility of results shown in HeLa cell DNA extraction using MagCore® systems. HeLa cell cultures in 2-fold serial dilutions were subjected to DNA extraction. All samples were treated with RNase A to remove RNA contaminants. Duplicates of samples with different amounts of cells were analyzed: 4 x 106 cells, 2 x 106 cells, 1 x 106 cells, and 5 x 105 cells. The yield and purity from samples of 200µl cultured cell suspensions were measured by Nanodrop n-1000. Lane 1/2: 4 x 106 Hela cells, Lane 3/4: 2 x 106 Hela cells; Lane 5/6: 1 x 106 cells; Lane 7/8: 5 x 105 cells and Lane M: RBC 1kb ladder marker.

    Table 2. Results of cell DNA extraction from amniotic fluid by Nanodrop n-1000. Cells were harvested from 10-15ml amniotic fluid from healthy females pregnant for 16-18 weeks. Purified by MagCore® instruments, the DNA yield varied between individuals due to different body conditions. However, the purity (A260/A280) were consistent in the range of 1.8 ±0.1.
